A preliminary study on the expression of CD44s in some skin tumors

Li Yuanhong, He Chundi, Chen Hongduo. Department of Dermatology, First Hospital of China Medical University, Shenyang, 110001

[Abstract]  Objective: To investigate the expression of CD44s in some skin tumors.  Methods: Immunohistochemical method was used to detect the expression of CD44s, V4/5 and V6 in squamous cell carcinoma (SCC), basal cell carcinoma (BCC), mycosis fungoides (MF), Bowen¡¯s disease, malignant melanoma (MM), seborrheic keratosis, melanocytic naevus and normal skin. Results: In SCC and BCC the cancer nest was more nearer to the epidemis the expression of CD44s was more stronger. The expression of CD44s in MM and naevus was similar to that in SCC and BCC. In other skin tumors the expression of CD44s was as the same as the normal skin.  Conclusion: The expression of CD44s was correlated with the location of the skin cancer nest and there is no association between the expression of CD44s and the malignancy of naevus cells.

The expression of vascular endothelial growth factor and the density of tumoral microvessels in non-invasive and invasive basal cell carcinoma

Deng Liehua, Luo Yuwu, Zhao Gang, et al. Department of Dermatology, The First Affliated Hospital of Medical College of Jinan University, Guangzhou, 510630

[Abstract]  Objective: To investigate the expression of vascular endothelial growth factor (VEGF), the density of tumoral microvessels (TMVD), and their significance in non-invasive basal cell carcinoma (NIBCC) and invasive basal cell carcinoma (IBCC).  Methods: We used SP immunohistochemical staining for observing VEGF and Von-Willebrand factor in NIBCC and IBCCs. Results: There were no statistically significant differences in the moderate-level expression of VEGF between NIBCC and IBCC (p=0.189). The level of TMVD in IBCC was higher Than that in NIBCC (p=0.019). The expression of VEGF was positively correlated both in NIBCC and IBCCs(rs1=0.667, p=0.003; rs2=0.582, p=0.014).  Conclusion: Based on this study VEGF might not be taken as an indicator of the malignancy in BCC, while TMVD is a good indicator of the malignancy in BCC, while TMVD is a good indicator in assessing the invasiveness of BCC.

Determination of serum SIL-2R level and its clinical significance in syphilitic patients

Lin Linlin, Wu Huizhen, Chen Yunqin, et al. Department of Dermatology, First Affiliated Hospital Wenzhou Medical college, Wenzhou, 325000

[Abstract]  Objective: To determine the serum SIL-2R level and its clinical significance in syphilitic patients.  Methods: The serum SIL-2R level and RPR change before and after benzylpenicillin therapy in patients with syphilis were determined with polyclonal antibody sandwich ELISA. The correlation of serum SIL-2R level with different stages of syphilis was analyzed.  Results: The serum SIL-2R level was higher in all stages of syphilis than that in control group (p<0.001) and was higher in both secondary and latent syphilis than that in primary syphilis (p<0.01). There was no significant difference in serum SIL-2R level between secondary and latent syphilis (p>0.05). The titer of serum SIL-2R and RPR decreased in all stages of syphilis after effective treatment. Compared with control group, the serum SIL-2R level in secondary and latent syphilis was still higher (p<0.05). Conclusion: The continual dynamic monitoring of SIL-2R level for the same patient could be used for evaluating the activity of illness and the therapeutic effect.

The impact of ceramide on differentiation of cultured keratinocytes

Sun Ling,  Zhao Ye. Department of Dermatology, Second Affiliated Hospital, Dalian Medical University, Dalian, 116023

[Abstract]  Objective: To investigate the impact of ceramide on the differentiation of keratinocytes cultured in serum-free medium.  Methods: Two kinds of ceramide (C2 and C6) were selected and added into serum-free media with keratinocyte culture. In 0,3,6,12 and 24 hour after adding ceramides, total RNA was abstracted to detect the expression of specific keratinocyte differentiation markers: TGasel, IVL, KRT1 KRT10, etc. by RT-PCR method.  Results: The expression of TGasel mRNA and IVL Mrna was most significant in 12 hours in the media containing C2 ceramide, and the levels were 5.7 times and 5.2 times than that in the control media without C2 ceramide in 12 and 24 hours, respectively. The expression of KRT1 Mrna and KRT10 Mrna in the media containing C2 ceramide was also increased but did not reach a significant level. The expression of Tgasel Mrna and IVL Mrna in the media containing C6 ceramide was similar to the media containing C2 ceramide, and was 3.3 times and 3.5 times than that in control media in 24 hours. However, the expression of KRT1 Mrna and KRT10 mRNA was significantly higher in the media containing C6 ceramide than in the media containing C2 ceramide, with 10 in the former and 5.4 times in the later. Conclusion: Ceramide could significantly increase the expression of specific differentiation markers (protein mRNA) of keratinocytes cultured in serum-free media and could induce the differentiation of keratinocyte.

Primary Vibrio vulnificus infection: A report of 12 cases

Li Bingxu, Wu Huizhen, Lu Zhongqiu, et al. First Hospital of Wenzhou Medical College, Wenzhou, 325000

[Abstract]  Objective: To analyze 12 cases with primary Vibrio vulnificus infection (PVVI).  Methods: Clinical profiles of 12 cases with primary PVVI were collected and analyzed from 1995 to 2000.  Results:  There were 8 males and 4 females. The average age was 48.6 years, ranging from 40 to 67 years. Six of the 12 patients had cirrhosis and the medical history of other 6 cases was nonspecific. The onset of the disease started suddenly with fever and occurred in late summer or early autumn in most cases. The most prominent clinical presentations were painful swollen, bulla and hemorrhagic bulla in lower extremities. Vibrio vulnificus was cultured positively in bullus fluid from 2 patients. The main pathological findings were local acantholysis, separation of epidemis from dermis, degeneration of collagen fibers, vasculitis and occlusion of small arteries. Under electromicroscopy, the microorganism appeared arc like with flagellum. Eight of the 12 cases died, 1 recovered and 3 lift the hospital voluntarily. The main cause of the death was multi-organ dysfunction. Conclusion: PVVI is an unrecognized acute disease with high death rate.
